Intertek has developed and implemented Direct Peptide Reactivity Assays (DPRA) to help determine the Skin Sensitization potential of new and/or existing ingredients used in cosmetics, various consumer and industrial finished products. Methods have been developed using model peptides standardised for groups of chemicals with varied reactivity reaction mechanisms, leading to Direct Peptide Reactivity Assays that demonstrate adduct formation. Intertek’s GLP accredited laboratories and experienced toxicology and safety assessment expertise provides data for your product’s potency profiles and safety assessment of ingredients whilst eliminating the need for animal testing.

Direct Peptide Depletion Studies

Intertek screen sets of chemicals which are brought into contact with specific choice of relevant nucleophiles (e.g. lysine, cysteine or glutathione) containing peptides. Using these assays we can determine both quantitative (disappearance of target peptide) and qualitative (observe the formation of covalent adducts to peptide aspects of chemical reactivity which combined provide a basis for predicting skin sensitisation potential. The formation of adducts is postulated as an indication of the potential allergenistic effect of the test compound. Sets of test chemicals can be fairly rapidly screened using an LC-MSMS approach.
